Mouse Plasmacytoma Growth In Vivo : Enhancement by Interleukin 6 ( IL - 6 ) and Inhibition by Antibodies Directed against II , - 6 or Its Receptor

چکیده
Murine plasmacytomas show a striking dependence on interleukin 6 (IL6) for their growth in vitro . Here, we present evidence suggesting that IL6 also plays an essential role in the in vivo development of these tumors . This conclusion is based on the finding that the tumorigenicity of an IL-6-dependent plasmacytoma cell line was increased -100-fold on transfection with an IL-6 expression vector, whereas it was inhibited in animals treated with monoclonal antibodies capable of blocking the binding of IL6 to its receptor. Injection of these antibodies 1 d before tumor challenge protected >50% ofthe mice and retarded tumor growth in all animals . Tumors arising in antibody-treated mice retained their 11,6 dependence in vitro, suggesting that the level of protection could be improved if stronger IL6 antagonists were available.

متن کاملT cell growth and differentiation induced by interleukin-HP1/IL-6, the murine hybridoma/plasmacytoma growth factor
Interleukin-HP1 (HP1)/IL-6 is a 25-30-kD protein produced by macrophages, fibroblasts, and certain T cell lines. It was originally identified as a mouse growth factor for B cell hybridomas and plasmacytomas, and was recently shown to stimulate growth and differentiation of normal B cells. Here we demonstrate that, in the presence of lectins or anti-T cell receptor antibodies, HP1/IL-6 has a gro...

We analyzed the stimulatory effect of oncostatin M (OSM), leukemia inhibitory factor (LIF), interleukin 6 (IL-6), IL-11, and the inhibitory effect of anti-IL-6 antibody (Ab), anti-IL-6 receptor monoclonal antibody (mAb), and anti-gp130 mAb on the growth of human plasmacytoma cells freshly isolated from a patient with multiple myeloma. The purified cells showed a plasmacytoid morphology and expr...

متن کاملInterleukin-6 family of cytokines and gp130.
G ROWTH AND DIFFERENTIATION of cells in the immune and hematopoietic systems are regulated by cytokines or interleukins. One of the characteristic features of cytokines is their functional pleiotropy and redundancy, ie, one cytokine shows a wide variety of biologic functions on various tissues and cells, and several different cytokines exert similar and overlapping functions on a certain cell. ...
